چکیده

Background: MicroRNA-99a has been believed to play a critical role in progression in various cancers; however, the prognostic and clinicopathological value of microRNA-99a in cancers remains unclarified. Methods and Results: We first evaluated the prognostic significance of microRNA-99a expression in 21 human cancer types in The Cancer Genome Atlas (TCGA). Patients with lower microRNA-99a levels had lower chances of overall survival in lung adenocarcinoma, esophageal cancer, cervical cancer, and head and neck squamous cell carcinoma, whereas they were more likely to survive urothelial bladder cancer and cutaneous melanoma. Subsequently, we included seven studies with sufficient data to further assesse microRNA-99a expression and its prognostic significance in human cancer types using meta-analysis. Low microRNA-99a level was closely linked with poor survival according to the published studies (HR=2.531, 95% CI: 1.920~3.337), but not in TCGA data (HR=1.042, 95% CI: 0.971-1.119). It clearly showed that down regulation of microRNA-99a was closely associated with shorter survival according to published studies and TCGA data (HR=1.101, 95% CIs 1.028~1.179). Finally, the bioinformatics analysis revealed that microRNA-99a might be involved in complex cellular pathways by Gene Ontology (GO) Biological Processes term, such as regulation of transcription. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that glycosaminoglycan biosynthesis and mitogen-activated protein kinase (MAPK) pathway were the most significant pathways regulated by microRNA-99a. The Search Tool for the Retrieval of Interacting Genes/ Proteins (STRING) also visualized the protein interaction and the related pathways in this study. Conclusions: These results indicate that microRNA-99a may be a reliable indicator for the progression and outcome in cancers patients.

Evaluation of MicroRNA-99a and MicroRNA-205 Expression Levels in Bladder Cancer

Bladder cancer is the second most common cancer in the genitourinary tract, showing often recurrence and progresse into invasive states. Epigenetic changes, such as microRNA alteration are involved in bladder cancer tumorigenesis through a variety of signaling pathways. The epigenetic state depends on geographic and lifestyle conditions. The aim of this study was to investigate the expression l...

نقش microRNA-21 در سرطان کولورکتال و قابلیت آن به عنوان یک بیومارکر پیش بینی کننده، تشخیصی و پیش آگهی: مقاله مروری

Colorectal cancer (CRC) is one of the most common types of cancers worldwide. Despite the improved therapeutic and diagnostic strategies and screening programs, morbidity and mortality of CRC is still considerable. Currently, most common approaches for diagnosis of CRC are colonoscopy and fecal occult blood test (FOBT). Because of the invasive nature of colonoscopy and low sensitivity of FOBT, ...

DNA methylation mediated silencing of microRNA-874 is a promising diagnosis and prognostic marker in breast cancer

MicroRNA-874 (miR-874) is downregulated in several human cancers and has been suggested to be a tumor suppressor gene. However, the molecular mechanism of miR-874 downregulation in breast cancer has not been well elucidated. Here we aimed to study the aberrant hyper-methylation of CpG sites with the utility of miR-874 downreregulation in breast cancer and evaluate the clinical function of miR-8...

A novel microRNA signature predicts survival in stomach adenocarcinoma

Recent microRNA (miRNA) expression profiling studies suggest the clinical use of miRNAs as potential prognostic biomarkers in various malignancies. In this study, aiming to identify microRNAs with prognostic value for overall survival (OS) in stomach adenocarcinoma (STAD) patients, we analyzed the miRNA expression profiles and the associated clinical characteristics in 380 STAD samples from The...

Identification of Prognostic Genes in Her2-enriched Breast Cancer by Gene Co-Expression Net-work Analysis

Introduction: HER2-enriched subtype of breast cancer has a worse prognosis than luminal subtypes. Recently, the discovery of targeted therapies in other groups of breast cancer has increased patient survival. The aim of this study was to identify genes that affect the overall survival of this group of patients based on a systems biology approach. Methods: Gene expression data and clinical infor...
